Azotobacter sp is used as a biofertilizer does nitrogen fixation by taking atmospheric nitrogen and converting it in to organic nitrogen and ammonia. The enzyme nitrogenase Coded by Nif gene cluster present in the genome of Azotobacter sp carries out nitrogen fixation. In this study, 95 samples were positive out of hundred marine samples were collected in different locations of Rameshwaram marine region (Gulf of Mannar) at the depth of 1-2m. The Azotobacter sp was isolated from the samples by the use of selective Azotobacter medium containing 3.5% sodium chloride. Further the chromosomal DNA was quantified by UV spectrophotometer. The nucleic acids (DNA) were isolated from the test Azotobacter sp and checked the presence and purity on 0.8% agarose gel electrophoresis and the bands were found to be uniform. PCR detection of the NifH primers have been successfully applied to describe diazotroph populations in the marine environment, but Selective NifH primer from Anabaena sp strain PCC7120 was used for the amplification of the Azotobacter sp the primers used in my study was exactly showing the result for Azotobacter genome. However, the use of highly degenerate primers and low-stringency amplification conditions render these methods prone to amplification bias, while less degenerate primer sets will not amplify all NifH genes.
